What is the black plastic square next to the rear grab handle?

It's a speaker for the infotainment system, well at least it is on mine. I have the upgraded B & O system. If you haven't got that system then maybe that's why it is blank
The thing circled on the door is a speaker (tweeter) but we're talking about the object on the headliner by the grab handle.

Generally speaking, the thing next to the grab handle over the door, is a cover that reveals a "receptable" (?) for a cargo restraining net. I'm not sure if it is a legal requirement here in Europe (I can check the EC regulations but it takes time), but a cargo net is a reasonably popular accessory.
I like the cargo mounting port explanation better than the speaker explanation. The keyhole slot seems meant for fastening something, and looks too small for a speaker to fit inside.

Could someone possibly take a good photo of it so it can be settled?

So, behind the "lid" or cover on my Euro Mach E, is the key shaped holes for a cargo net. Both in the back seat and in the trunk area.

A quick search in the EC regulations found no demand for these but they are in "every" European Ford I have had the last 20 years, as far as I can remember.

Speakers for the B&O system must be different between Europe and NA. I wonder why?

Why in the back seat? For when you fold the seats to use your Mach-E to the fullest :)

But of course here it is still in the trunk.
Oh! To keep pets from becoming “meat-and-bone” missiles in the event of an accident! Cool!

My 2018 Volvo XC60 had something similar, though it lacked the cover if I recall correctly... It was definitely for a doggie net. I never got one though, because cats.

Gotta say, this thread could have benefitted from some specificity from the beginning. Anyone know what that grey circle thing is over there? 😂
There's a thing under the part in the front that makes that clicky noise. Do you know what the tab on the back behind the coverplate with the two notches is? And how can I get it to beep, or whistle, instead of click? Ford never should have called this thing a Mustang...
